诱细胞衰老,促免疫清除:南京中医药大学邵江娟/郑仕中/程海波团队揭示中药黄芩活性成分千层纸素A改善肝纤维化的全新双重机制

肝纤维化是慢性肝病走向肝硬化和肝癌的关键转折点,目前全球缺乏有效的抗纤维化药物。肝星状细胞(HSCs)的持续活化是推动纤维化的核心环节,如何有效清除活化的HSCs是治疗的重大难点。近日南京中医药大学邵江娟/郑仕中/程海波团队揭示了一种双重调节机制:千层纸素A(Oroxylin A,OA)对SIRT7的抑制能够重编程HSCs命运:PRMT5琥珀酰化驱动的HSCs衰老和外钙网蛋白(ecto-CRT)依赖的NK细胞免疫清除。这项工作确立了SIRT7作为关键的治疗靶标,并为开发抗肝纤维化策略提供了新见解。相关成果以“Dual Roles of SIRT7 Inhibition by Oroxylin A Reprogram HSCs Fate: PRMT5 Succinylation-Driven Senescence and Ecto-Calreticulin-Dependent NK Cell Immune Clearance in Liver Fibrosis”为题发表在Research上。
