High-dose taurine treatment exacerbated liver damage and hepatic fat accumulation in an ALD mouse model. (IMAGE)

(A) Schematic illustration of experimental design. Mice were fed normal chow diet with water containing taurine at 3 g/kg (calculated based on daily drinking amount) for 10 days. Mice were then subjected to the NIAAA chronic-plus-binge alcohol feeding in a liquid diet containing 0–5% EtOH (v/v) ramping up in 6 days and remaining at 5% EtOH for 10 days, with taurine added to the liquid diet. On the last day, the mice were orally gavaged with 5 g/kg EtOH 9 hours before euthanising. PF mice were treated isocalorically with maltose dextrin and similarly orally gavaged with maltose dextrin 9 hours before euthanising. (B) Liver-to-body weight ratios (%). (C) Hepatic triglyceride content (mg/g liver tissue). (D) Representative H&E-staining of liver sections (upper panel: ×10 magnification; lower: ×20 magnification). (E) Serum ALT and AST levels. (F) TUNEL staining of liver sections (×20 magnification, red arrows indicate positive staining). Data are presented as mean±SEM (n=4–10). *p<0.05, **p<0.01, ***p<0.001, ****p<0.0001. AF, alcohol-fed group; AF+3T, alcohol-fed group supplemented with 3 g/kg taurine; ALD, alcohol-associated liver disease; ALT, alanine aminotransferase; AST, aspartate aminotransferase; EtOH, ethanol; NIAAA, National Institute on Alcohol Abuse and Alcoholism; PF, pair-fed control group; PF+3T, pair-fed group supplemented with 3 g/kg taurine; taurine, β-aminoethanesulfonic acid; TUNEL, terminal deoxynucleotidyl transferase–mediated dUTP Nick-End Labeling.
