Schematic overview of the obtained results. Bile cfDNA from patients with biliary strictures obtained at the first ERCP was analysed by ULP-WGS and NGS of 52 targeted genes (Bilemut assay). ULP-WGS enables early malignancy detection with high specificity and tumour-origin discrimination through TF and CNA profile. Integration of TF with the targeted TMB identifies a subgroup of patients with poorer prognosis. Figure created using BioRender. com, CCA, cholangiocarcinoma; cfDNA, cell-free DNA; ERCP, endoscopic retrograde cholangiopancreatography; NGS, next-generation sequencing; PDAC, pancreatic ductal adenocarcinoma; ULP-WGS, ultra-low-pass whole-genome sequencing.
