AC004854.2 enhances tumorigenesis in colorectal cancer xenografts through c-MYC-dependent mechanisms. (IMAGE)
Caption
(A) Xenograft models were established by subcutaneous injection of HCT116 cells overexpressing AC004854.2 or empty vector control, followed by intraperitoneal administration of the c-MYC inhibitor IZCZ-3 or saline control. (B) Tumor volume was measured every three days after implantation, and statistical analyses were initiated on day 12. (C) Tumors were harvested at endpoint following euthanasia. (D) Tumor weight was determined ex vivo. (E) AC004854.2 expression levels in xenograft tissues were validated by quantitative reverse transcription PCR. (F) Immunohistochemical staining was used to detect protein expression of key proliferation and metastasis-associated markers, including Ki-67, CCND2, CDK4, E-cadherin, and c-MYC. (G, H) The mRNA (G) and protein (H) levels of PUF60, FUBP1, c-MYC, CCND2, and CDK4 were assessed by quantitative reverse transcription PCR and Western blotting analysis, respectively. Statistical analyses: Wilcoxon rank-sum test, Student's t-test; significance levels: ns (not significant); ∗∗∗P < 0.001.
