Experiment-guided AlphaFold produces models in agreement with measurements, even when AlphaFold3 completely mispredicts key structural features. Reconstruction of an amyloid-beta fibril using cryoEM electrostatic potential (ESP) map guidance (green). AlphaFold3 completely mispredicts the dimerization interface and the structure of the dimeric unit. ESP guidance produces accurate agreement with the electron density map.
