A spatiotemporal single-cell atlas of 120,649 immune cells across 10 tissues identifies PDCD4 as a microbiota-responsive brake on CD8+ T cell activation (IMAGE)
Caption
A total of 120,649 CD45+ immune cells were sorted from 10 tissues of germ-free (GF) and specific pathogen-free (SPF) mice at postnatal day 16, week 4, and week 9, and profiled by MARS-seq single-cell RNA sequencing. Microbiota exposure selectively expanded specific myeloid and lymphoid clusters, maintain antigen-processing capacity in developing myeloid lineages, and relieve PDCD4-mediated translational repression in lymphocytes.
