Context-dependent RNA m5C regulation and cancer-associated phenotypes. (IMAGE)
Caption
Mammalian NSUN1–NSUN7 and DNMT2/TRDMT1 install m5C in different RNA classes, whereas ALYREF and YBX1 recognize selected methylated transcripts and influence RNA processing, nuclear export, transcript stability, and translation. The oncological examples shown are illustrative rather than components of a universal regulatory pathway. NSUN2/YBX1-mediated regulation of QSOX1 has been linked to gefitinib resistance, whereas an NSUN5/TET2-dependent oxidation pathway has been associated with immune evasion in glioma. The figure summarizes biological plausibility and does not imply analytical or clinical validation of an RNA m5C biomarker. ALYREF: Aly/REF export factor; C: cytosine; DNMT2: DNA methyltransferase 2; m5C: 5-methylcytosine; mRNA: messenger RNA; NSUN: NOP2/Sun RNA methyltransferase family; QSOX1: quiescin sulfhydryl oxidase 1; RNA: ribonucleic acid; TET2: tet methylcytosine dioxygenase 2; TRDMT1: transfer RNA aspartic acid methyltransferase 1; YBX1: Y-box binding protein 1
