(a) ATM inhibitors protect MDA-MB-231 cells against ferroptosis. MDA-MB-231 cells were treated with increasing doses of erastin (24 h), either alone or in combination with liproxstatin-1 (LPT1, 2 μM), KU55933 (5 μM), and KU60019 (1 μM). Cell viability was quantified by CellTiter-Glo assay; (b, c) AZD1390 reduced erastin-induced ferroptosis in MDA-MB-231 cells in a dose-dependent manner. After 24 hours of incubation, cell viability and cytotoxicity were determined by CellTiter-Glo assay (b) and CellTox Green assay (c), respectively. Canonical ferroptosis inhibitors: Fer-1; DFO; (d, e) Erastin-induced lipid peroxidation (10 μM, 18 hours) in MDA-MB-231 cells was reduced by AZD1390; Lipid peroxidation was assessed by C11-BODIPY staining (d) and quantified as the percentage of lipid peroxidation-positive cells (e). (a, b, and e) Data represent the mean ± SEM. (a, b) 2-way ANOVA and Dunnett’s multiple comparisons. (e) One-way ANOVA followed by Dunnett’s multiple comparison test. (c) Non-parametric Mann-Whitney tests with correction for multiple comparisons. Data represents the median ± IQR. (a-e) n = 6 independent biological replicates. *p < 0.05; **p < 0.01; ***p < 0.001, ****p < 0.0001. MDA: malondialdehyde; ATM: ataxia-telangiectasia mutated; LPT1: liproxstatin-1; Fer-1: Ferrostatin-1; DFO: Deferoxamine; SEM: standard error of the mean; ANOVA: analysis of variance; IQR: interquartile range.
