KAIST Identifies a “Hidden Amplification Switch” in Lymphangiogenic Signaling, with Implications for Lymphedema and Cancer Metastasis (IMAGE)
Caption
Figure 1. Cluster formation of the VEGFR-3/VEGF-C complex and signal amplification
- Cryo-EM observation of the VEGFR-3/VEGF-C complex showed that multiple complexes assemble into two distinct types of higher-order clusters.
- The three-dimensional structure of the cis-cluster, in which multiple complexes are aligned side by side, was determined.
When VEGFR-3 was mutated to disrupt cis-cluster formation, the activation of VEGFR-3 and ERK was markedly reduced despite VEGF-C stimulation. This confirms that cis-clustering is critical for signal amplification.
