Dietary lipids set the threshold for of T-cell ferroptosis (IMAGE)
Caption
In mice and healthy human cohorts, a high systemic PUFA/MUFA ratio favors ACSL4-dependent incorporation of PUFA into T-cell phospholipids, increasing lipid peroxidation and ferroptotic stress. This weakens T follicular helper (Tfh) cell cell-supported humoral immunity and limits the persistence and effector activity of tumour-reactive T cells, including chimeric antigen receptor (CAR)-T cells. A lower PUFA/MUFA ratio has the opposite effect, promoting a membrane environment that is less permissive to peroxidation and more supportive of adaptive immune function.
