TREM2-Candidalysin-DAP12/Syk/NF-κB-TNF-α Axis (IMAGE)

Upon recognition of candidalysin, TREM2 recruits the adaptor protein DAP12 to activate Syk kinase, subsequently phosphorylate NF-κB p65 and MAPK p38, and ultimately lead to the secretion of TNF-α. TNF-α acts in an autocrine manner to enhance macrophage phagocytosis, killing, and reactive oxygen species production. TNF-α also promotes neutrophil extracellular trap (NET) formation and bactericidal function, while moderately modulating IL-17 signaling from TCRαβ+ cells in a paracrine manner.
