Utilizing large-scale transsynaptic tracing, Cai et al. reconstructed 3D space of 50,168 cerebellar nuclei (CN)-projecting neurons in the thalamus, midbrain, and brainstem. They reveal significant differences in the projections to target nuclei due to neuloligin3R451C mutation. Selective inhibition of the CN→zona incerta pathway can ameliorate social deficits in neuloligin3R451C mice.
